一次运行多种pH范围的IPG胶条，获得最大限度的分离效果，数据重现性高
PROTEAN i12 IEF系统可以在一次运行中，使用不同pH范围的IPG胶条，从而获得最大限度的分离效果。选择pH3-10的宽范围胶条，比较其与pH3-6，pH5-8，pH7-1三种窄范围胶条重叠所获得的2D图，四种胶条同时运行，一次便可获得所有数据。红圈内突出显示了窄范围胶条的高分辨率，表格内显示了三条重复胶条内检测到的蛋白点数，说明重复性非常好。
来自小鼠肝脏的样品通过尿素-硫尿-CHAPS溶液提取。提取液分别用12条不同pH范围，长度均为11 cm的ReadyStripTM IPG胶条进行等电聚焦试验。所有的胶条都放置在一台PROTEAN i12 IEF仪器的聚焦盘内，同时运行。IPG胶条的pH范围分别为：3-10，3-6，5-8和7-10，每种pH范围重复3次。第二向SDS-PAGE试验使用浓度为8-16%的梯度CriterionTM预制胶，然后用考马斯蓝染色。上图为使用不同pH范围的lPG胶条最后获得的2-D电泳图和三个重复样本所获得的蛋白点数。表格显示三个重复胶条中都存在的蛋白点数。
